Equipment
1. 量筒、烧杯、耐高温高压蓝盖玻璃瓶（Thermo fisher)；
2. Rapid-Flower Filter（滤器）（Thermo Nalgene）。
Procedure
以配制1L 10×PBS储存液为例：
1、 用双蒸水清洗量筒、烧杯、10×PBS储存液玻璃瓶（3次）。
2、 使用一次性称量船，分别用分析天平称取Na2HPO4.12H2O，35.814g；KH2PO4，2.4496g；NaCl，80.0669g；KCl，2.0129g。
3、 将药品粉末倒入PBS配制专用烧杯中，加入800-900ml miniQ水，使用磁力搅拌器充分溶解（无任何固体）。
4、 用移液枪吸取2-3ul溶液滴在pH试纸上测试酸度，pH应为6.8。
5、 将溶液转移到PBS专用量筒或容量瓶中定容到1L（加miniQ水至1L），倒回烧杯中充分混合。
6、 将滤器与微型台式真空泵相连，打开微型台式真空泵后，将PBS分批倒入滤器进行过滤。滤器价格昂贵，建议一次过滤4-5L溶液。
7、 滤好的10XPBS可立刻稀释使用；稀释后可用pH试纸再次确认pH值。
8、 暂时不用的10XPBS分装至10×PBS储存液玻璃瓶中（液面不超过瓶身的3/4高度），高压灭菌。 注意事项：瓶盖不能完全拧紧（保留约半圈），用锡箔纸包裹瓶盖；不能放到底层篮子，容易被脏水污染；降温后一旦打开灭菌锅，必须立刻拧紧盖子，防止外界细菌进入。
9、 灭菌后的10XPBS可保存在室温或4℃冰箱中。每次分装后需盖紧瓶盖，降低污染概率。
10、取用前注意观察瓶底有无沉淀，摇晃后有无悬浮物；若有说明有微生物滋生，请勿使用，需重新配置。
11、每配500ml1XPBS可用50ml进口管测量50ml 10XPBS，加入450ml miniQ水，颠倒混匀。

10×PBS储液的pH是6.8，稀释为1×PBS时，pH变为7.4。
